Infiammazione e risposte del sistema immunitario (Teresa ZELANTE)
    My PhD was based on the study of the immunology of opportunistic diseases. In particular, in understanding the role of the Th17 cell response in early and late phases of infection. At the Singapore Immunology Network, I had my postdoc where I investigated the role of IL-2 in innate immune cells.

    Directly under the Program Research ‘Giovani Ricercatori Rita Levi Montalcini’, I have found that microbes in the lung may activate a pathogenic response AhR-mediated. Therefore, in my research programs, I had the chance to develop expertise in Microbiology and Immunology. In the last years, I have focused on mouth/lung/gut microbiome studies by using as interface the 3D organoid model.

    We found that the Th17 pathway dampens tryptophan catabolism, and thus may favour pathology and accommodate the seemingly paradoxical association of chronic inflammation in fungal diseases. We also studied whether the cytokine IL-17A/F affected Candida gene transcription. Here, we found that chronic exposure to IL-17A/F affected Candida survival and germination rate, whereas short exposure induced the autophagic machinery response. We also found that a membrane protein bound the cytokine and was responsible for the intracellular response of Candida to IL-17A.

    We are focused in understanding the oral microbiome in vivo systems of oral mucositis as well as in vitro systems by using oral tongue organoids. Our main aim is to provide novel therapies for oral mucositis in patients ndergoing radiotherapy

    Studies focused on the microbe-host interaction based on more than 20 years of research experience in mechanisms of interaction between the host immune system and fungi as opportunistic pathogens. In particular, almost ten years ago, we have found that the mammalian enzyme IDO helps in regulating the homeostasis of the gut microbiome. Increased availability of tryptophan in vivo (simulated by using IDO-deficient mice) perturbs the system by favouring survival and proliferation of Lactobacilli, in particular L. reuteri, which is able to metabolize tryptophan and release secondary products that bind the AhR

    Developing of organoids from healthy and cancer stem cells. The technology is extended on various tissues and biopsies and it has been founded recently by internal resources.
